INTRODUCTION ANNUAL BLUEGRASS
Annual Bluegrass (Poa annua) is a monocot weed in the Poaceae family.  In Tennessee this weed first evolved multiple resistance (to 7 herbicide sites of action) in 2017 and infests Golf courses.  Multiple resistance has evolved to herbicides in the Groups 2 (Legacy B), PSII inhibitors - Serine 264 Binders HRAC Group 5 (Legacy C1 C2), Inhibition of Protoporphyrinogen Oxidase HRAC Group 14 (Legacy E), Inhibition of Enolpyruvyl Shikimate Phosphate Synthase HRAC Group 9 (Legacy G), Inhibition of Glutamine Synthetase HRAC Group 10 (Legacy H), Inhibition of Microtubule Assembly HRAC Group 3 (Legacy K1), and Inhibition of Cellulose Synthesis HRAC Group 29 (Legacy L).  These particular biotypes are known to have resistance to flumioxazin, foramsulfuron, glufosinate-ammonium, glyphosate, Indaziflam, metribuzin, prodiamine, propyzamide/pronamide, and simazine and they may be cross-resistant to other herbicides in the Groups 2 (Legacy B), PSII inhibitors - Serine 264 Binders HRAC Group 5 (Legacy C1 C2), Inhibition of Protoporphyrinogen Oxidase HRAC Group 14 (Legacy E), Inhibition of Enolpyruvyl Shikimate Phosphate Synthase HRAC Group 9 (Legacy G), Inhibition of Glutamine Synthetase HRAC Group 10 (Legacy H), Inhibition of Microtubule Assembly HRAC Group 3 (Legacy K1), and Inhibition of Cellulose Synthesis HRAC Group 29 (Legacy L).

Herbicide resistance within Poa annua is widespread in managed turfgrass systems. In 2020, a P. annua collection from a golf course in the southeastern United States was reported to be resistant to indaziflam as well as six other mode-of-action groups. This first report in 2020 suggests that turfgrass managers would benefit from a bioassay to screen other collections with putative indaziflam resistance. A dose-response experiment was conducted with ten concentrations of indaziflam (0, 250, 500, 667, 1000, 1143, 1333, 2000, 4500, and 9000 pM) in Gelrite® culture during 2021 and 2022. An herbicide-susceptible (S1) collection of P. annua, a resistant standard (Site 3A), and a collection with putative resistance to indaziflam (Site 18) were included in this experiment. Petri dishes were filled with 80 mL of Gelrite® (3.75 g L−1) containing technical grade (≥ 98%) indaziflam and rifampicin (1000 µg mL−1). Each plate was sealed with parafilm after placing 15 seeds of a single collection on the Gelrite® surface. At 14 days after seeding (DAS), the length of the radicle (mm) protruding from each seed was recorded with digital calipers. Indaziflam concentrations required to reduce root growth by 70% (EC70) were calculated via non-linear regression. Statistically significant differences were detected among P. annua collections with the EC70 values for the herbicide-susceptible collection measuring 708 pM [95% confidence interval (CI) = 656 to 764 pM] compared to 2130 pM (CI = 1770 to 2644 pM) for Site 3A and 4280 pM (CI = 3464 to 5442) for Site 18. Given that resistant collections exhibited longer root length in the absence of herbicide, confocal microscopy analysis was used to explore differences in root cell count among resistant and susceptible P. annua collections; however, few differences in cell count were detected. Overall, these findings indicate that a discriminatory dose of 708 pM (95% CI = 656 to 764 pM) can be used to differentiate among susceptible and resistant P. annua collections from field sites where poor control is observed following broadcast applications of indaziflam.

Annual bluegrass (Poa annua) resistance to indaziflam applied early-postemergence

Abstract
BACKGROUND
Indaziflam is an alkylazine herbicide used to control annual bluegrass (Poa annua L.). Several locations in the southeastern USA reported poor annual bluegrass control following treatment with indaziflam during autumn 2015. A series of controlled environment experiments were conducted to confirm putative resistance to indaziflam in annual bluegrass collected from these field locations.
RESULTS
Indaziflam (25 g ha−1) effectively controlled all putative-resistant annual bluegrass collections when applied preemergence (PRE), but was ineffective when applied early-postemergence (< 2.5 cm plant height; BBCH scale = 1; EPOST). In agarose-based plate assays, EPOST I50 values for putative-resistant collections ranged from 2424 to 4305 pm compared with 633 pm for the herbicide-susceptible control; therefore, resistance indexes (R/S) ranged from 3.8 to 6.8. Resistant collections were not controlled by foramsulfuron, flumioxazin, glyphosate, glufosinate, metribuzin, pronamide, or simazine applied EPOST. Indaziflam content in herbicide-susceptible annual bluegrass was greater than a resistant collection from 0 to 10 days after treatment (DAT). Susceptibility was not restored when resistant collections were treated with indaziflam plus 1-aminobenzotriazole (10 mg L−1), tebuconazole (1510 g ha−1), or malathion (400 g ha−1).
CONCLUSIONS
This is a first report of resistance to indaziflam in any plant. Additionally, we confirm that these annual bluegrass collections are resistant to several other herbicidal modes-of-action. It is unclear if this multi-herbicide resistance is due to a single resistance gene, multiple resistance genes, non-target site mechanisms, or a combination thereof. Additional research to better understand resistance mechanisms in these annual bluegrass collections is warranted.
